Overview
Thymosin β4 is a member of the thymosin β family. Thymosin β4 is composed of 43 amino acids and is hydrophilic.

Application prospects
The latest research shows that the expression level of acetic acid thymic β4 gene is positively correlated with cancer cell metastasis and is closely related to tumor apoptosis. This provides new ideas for finding new methods to treat cancer, and therefore has broad prospects for the development of new drugs.

Biological functions and mechanisms of action
Tβ4 is one of the main actin regulatory molecules in the human body. It has multiple biological functions and plays an extremely important role in physiological and pathological processes such as tissue regeneration, remodeling, wound healing, maintaining actin balance, tumor onset and metastasis, cell apoptosis, inflammation, angiogenesis, and hair follicle development. 1. Actin regulatory factor Actin accounts for about 10% of the total protein in non-muscle cells and is an important component required for cell structure, cell movement and wound healing. Tβ4 in cells is sufficient to block all actin and can participate in regulating the polymerization and depolymerization of actin. Tβ4 can bind to actin monomers at a ratio of 1:1 to prevent the formation of F-actin polymers. The binding of Tβ4 to actin is also accompanied by the dissociation of bound water. Its C-terminus binds to actin His-40, causing the conformation of actin monomers to change. The Tβ4 molecule contains an actin binding domain (LKKTET), which is the main electrostatic contact site. Its N-terminal pro-fragment can inhibit the polymerization of actin through steric hindrance. When acting alone, Tβ4 can inhibit the polymerization of actin and the exchange of nucleosides on actin. This is contrary to the effect of another actin binding protein Profiling, which can promote the exchange of ADP and ATP and accelerate the assembly of actin. Tβ4 and Profiling can synergistically regulate the assembly of actin. Tβ4 can regulate the conversion between G-actin and F-actin. It is reported that in highly mobile blood cells, the concentration of Tβ4 reaches 300pmol, while the concentration required for Tβ4 to bind to G-actin is less than 20pmol; when the concentration increases, Tβ4 reduces the ability of F-actin to depolymerize, which may be the reason why Tβ4 has the function of regulating the intracellular microfilament system.

Thymus β4 and tumors Studies have shown that thymus β4 acetate is associated with a variety of tumors, including colon cancer, melanoma, fibrosarcoma, breast cancer, pancreatic cancer and lung cancer, and it can cooperate with multiple factors to promote tumor invasion and metastasis. 2. The role of thymus β4 in the repair of cardiac damage In the acute stage of myocardial infarction, a large number of myocardial cells are ischemic and necrotic, resulting in impaired cardiac function and eventually progressing to heart failure. Studies have confirmed that thymus β4 acetate has the effect of protecting myocardial cells.
